CJC-1295 (no DAC / Mod GRF 1-29)
Modified GRF (1-29)
CJC-1295 (Mod GRF 1-29) Research Protocol
Quickstart Overview
CJC-1295 is a tetra-substituted analog of GHRH (1-29) engineered for resistance to DPP-IV degradation. The no-DAC form (Mod GRF 1-29) produces a sharp, physiological GH pulse; the DAC form binds albumin for a 6–8 day half-life.
Scientific Data
Dosing & Reconstitution Guide
Units assume a U-100 insulin syringe (100 units = 1 mL). Mod GRF 1-29 degrades faster in solution than most peptides — use within 2–3 weeks.
Reconstitution Steps
- 1Bring vial to room temperature; swab stoppers.
- 2Add 2.0 mL bacteriostatic water slowly down the glass.
- 3Swirl gently; solution should be completely clear.
- 4Label and refrigerate immediately.
Supplies Needed
Planning example for a 12-week no-DAC protocol at 100 mcg twice daily, paired with Ipamorelin.

Storage Instructions
Important Notes
- The DAC (Drug Affinity Complex) version produces continuous GH elevation ("GH bleed") rather than pulses — a different research model entirely.
- Saturation occurs ≈ 100 mcg; doubling the dose does not double the response.
- Extremely sensitive to improper storage — degraded Mod GRF loses efficacy rapidly.
- Monitor IGF-1 and fasting glucose in extended protocols.
How This Works
Four amino-acid substitutions (D-Ala2, Gln8, Ala15, Leu27) protect the GHRH (1-29) backbone from DPP-IV cleavage and oxidation, extending its half-life from minutes to ~30 minutes (no DAC). It binds GHRH receptors on somatotroph cells, elevating cAMP and triggering GH synthesis and release. Because it acts on the GHRH pathway rather than the ghrelin pathway, it is additive with GHS-R agonists like Ipamorelin. The DAC variant adds a maleimidoproprionic acid linker that covalently binds serum albumin, extending half-life to approximately 6–8 days.
Potential Benefits & Side Effects
- Amplified GH pulse amplitude in a physiological pattern
- Elevated IGF-1 sustained for days (DAC form)
- Improved body composition in animal studies
- Enhanced sleep quality and recovery markers
- Synergy with Ipamorelin — the most-cited secretagogue stack
- Flushing and head rush immediately after administration
- Water retention, especially with DAC form
- Injection-site redness or welts
- Headache and dizziness
- Carpal-tunnel-like symptoms at high doses
Lifestyle Factors
- Fasted timing is non-negotiable for GHRH analogs
- Avoid high-glycemic meals near doses; somatostatin surges blunt effect
- Sleep hygiene amplifies the nightly GH pulse
Injection Technique
- 1Subcutaneous abdominal; may be drawn into the same syringe as Ipamorelin.
- 2Inject slowly; this analog can produce transient site redness.
- 3Rotate sites.
